Challenges with complex flow paths, inconsistent droplet in PCR?

A life science customer set out to develop a DNA sample handling and preparation system capable of generating precise droplets of PCR oil-based reagent loaded with DNA. These droplets needed to be dispensed into well plates and seamlessly transferred to a digital PCR system for replication.

The challenge lay in designing a compact, integrated unit that could interface directly with disposable reagent bottles, manage complex fluid paths, and deliver consistent droplet formation—all while maintaining high throughput and reliability.

IMI’s Solution: Integrated Fluidics Manifold for Seamless Dispensing

IMI partnered with the customer to co-develop a customized fluidic assembly built around a 5-layer acrylic manifold. This innovative solution featured:

  • Chipsol On/Off valves, Media Separated valves, and a FLATPROP actuator for precise control.

  • Integrated sensors, fittings, and PCBs for real-time monitoring and automation.

  • Dual pressure chambers enabling an air-over-liquid system to extract reagent and direct it to a dedicated dispense head.

  • Direct bottle-to-manifold interface for simplified reagent loading.

This assembly allowed the dispense head to auto-fill well plates with high accuracy, reducing manual intervention and ensuring consistent droplet formation.
